Plasma membrane Ca2+-ATPases are responsible for the efflux of calcium ions across the plasma membrane and hence maintaining low levels of resting intracellular free Ca2+. Four genes encode for plasma membrane Ca2+-ATPases (PMCA 1-4). Increases in the expression of the plasma membrane calcium pump isoform 4 (PMCA4) occur in HT-29 colon cancer cell lines after sodium butyrate and post-confluency-induced differentiation, with no similar change in the other isoform expressed in HT-29 cells, PMCA1. Some clinical colon cancer samples are characterized by reduced levels of plasma membrane calcium pump mRNA suggesting that the less differentiated cells have compromised calcium efflux. Studies assessing the effects of PMCA4 overexpression and PMCA4 siRNA in HT-29 cancer cells suggest that the remodeling of calcium efflux in colon cancer cells does not increase sensitivity to apoptotic stimuli but instead may promote pathways important in the invasiveness and proliferation of colon cancer cells, through alterations in cell signaling and gene transcription, in particular that of early response genes
